Creative Enzymes performs a substrate titration on your restriction enzyme of interest and then analyzes it using mapping software to determine Km. Once Km is determined, that substrate concentration can be used in the inhibitor screening assay.
To test for solvent interference, Creative Enzymes titrates the solvent with the enzyme, looking for any increase or decrease in enzyme activity. If there is an interference, we determine the solvent concentration at which the interference becomes negligible. This solvent concentration is then used for the inhibitor screening test.
The inhibitor titrated in the solvent is incubated with the enzyme of interest. After incubation with the inhibitor, the activity of the enzyme is measured using the Km of the substrate concentration. Graphing software was used to plot the inhibition data and to determine the IC50 of the inhibitor.
