Factors Affecting Enzyme Stability
pH and buffers. Most enzymes are stable only within a specific pH range, beyond which stability is reduced.
Enzyme concentration. Although enzyme stability varies by nature and purity, in general, enzyme proteins are stable at high concentrations and are prone to dissociation and adsorption at low concentrations, and even to surface denaturation.
Some enzymes are sulfhydryl enzymes, which will be gradually inactivated by sulfhydryl oxidation in air. In this case, the addition of 1 mmol/L EDFA or DTT helps to improve their stability.
