DNA modifying enzymes play an important role in DNA replication, repair and recombination. Understanding the interactions between these enzymes and other molecules is essential for drug discovery, genetic engineering and biological research. At Creative Enzymes, we offer DNA modifying enzyme interaction assays to help you study the interactions between your target enzymes and various compounds or proteins.
Sepecific Assay Service We Offer
Our DNA modifying enzyme interaction assay service is designed to determine the binding affinity of your target enzyme to other molecules including:
DNA modifying enzymes with small molecules
DNA modifying enzymes with peptides
DNA modifying enzymes with nucleic acids
DNA modifying enzymes and proteins
By measuring binding affinity, we can help you identify interactions that are critical to the enzymatic activity of your target enzyme and also screen for potential enzyme inhibitors or activators.
Methodology
Surface Plasmon Resonance (SPR): SPR is a label-free, real-time technique that allows us to immobilize your target enzyme on a sensor chip and monitor the binding kinetics, affinity and specificity of the enzyme to different compounds or proteins.
Isothermal Titration Calorimetry (ITC): ITC is a highly sensitive calorimetric technique that measures the amount of heat released or absorbed when two molecules bind to each other.
Fluorescence Polarization (FP): We can use FP to measure the binding affinity and kinetics of interactions between your target enzyme and various compounds or proteins.
Electrophoretic Mobility Shift Assay (EMSA): EMSA is a gel electrophoresis-based technique that measures the change in mobility of DNA fragments upon binding to their interacting proteins, including DNA-modifying enzymes.
Sample Requirements
We require a minimum of 1 μg of purified enzyme or cell lysate per assay. We recommend that samples be frozen at -80°C until shipment.
Applications
By understanding the interactions between your target enzyme and other molecules, we can help you identify potential drug targets or develop new drugs that modulate the activity of the enzyme.
We can also screen for potential inhibitors or activators of enzymes and study the mechanism of action of these compounds.
In addition, our assay services can be used to study the functional role of these enzymes in DNA replication, repair and recombination, and to investigate the molecular basis of genetic diseases caused by mutations in these enzymes.
Deliverables
We provide our clients with detailed reports that include all experimental procedures and results obtained. The report includes a summary of the assay design, optimization, validation, and data analysis. We also provide an interpretation of the results and suggest further experiments if necessary.
