Tris-Glycine (hereinafter referred to as TG) is white to off-white instant granules. Each bag of TG instant granules can be formulated with 1 L of 1× TG buffer, which is easy to operate and easy to use. The main components of TG are Tris and glycine. The concentration of Tris in 1× TG buffer is 25 mM and the concentration of glycine is 192 mM.
1. The TG buffer is formulated into 1 × in 20% methanol and is often used for rapid western blotting (Western Blot), which is mainly used for wet transfer;
2. It is used to prepare non-denatured protein polyacrylamide gel (Native-PAGE) electrophoresis buffer.
Protocol :
1. Measure about 600 ml of distilled water into the beaker(if used for transfer in western blotting, 200 ml methanol and about 400 ml distilled water should be added), and place a magnetic stir bar in the beaker.
2. Place the beaker on a magnetic stirrer, slowly add the entire contents of 1 bag of TG instant granules, and stir the solution until it is completely dissolved.
3. Add distilled water to the TG solution in step 2, and dilute to 1 L, which is 1× buffer
After being formulated into 1× solution, the pH value is 8.30±0.20@25℃.
