TBE is white to almost white instant granules. Each bag of TBE instant granules can produce 1 L of 1× TBE buffer, which is easy to operate and easy to use. The main components of TBE are Tris, boric acid and EDTA. The concentration of Tris in 1×TBE buffer is 89 mM, the concentration of boric acid is 89 mM, and the concentration of EDTA is 2 mM.
TBE is a commonly used nucleic acid electrophoresis buffer in biology, mainly used for agarose gel electrophoresis of DNA. TBE has a strong buffering capacity, and it has a better separation effect when used for electrophoresis of fragments less than 1 kb, and it is also suitable for longer electrophoresis. TBE is easy to cause high electroosmosis when used in agarose gel, and the recovery rate of DNA fragments is reduced due to the interaction with agarose to generate non-covalently bound tetrahydroxyborate complexes, so it is not recommended for DNA Fragment recovery.
Protocol :
1. Measure about 600 ml of distilled water into the beaker, and place a magnetic stir bar in the beaker.
2. Place the beaker on a magnetic stirrer, slowly add the entire contents of 1 bag of TBE instant granules, and stir the solution until it is completely dissolved.
3. Add distilled water to the TBE solution in step 2, and dilute to 1 L, which is 1× buffer.
After being formulated into 1× solution, the pH value is 8.35±0.15@25℃.
*The concentration of TBE working solution usually used is 0.5×. In this case, 1× working solution can be diluted before use
