TAE is white to almost white instant granules. Each bag of TAE instant granules can produce 1 L 1× TAE buffer, which is easy to operate and easy to use. The main components of TAE buffer are Tris-acetate and EDTA-2Na. The concentration of Tris-acetate in 1×TAE buffer is 40 mM, and the concentration of EDTA 2Na is 1 mM.
TAE is a nucleic acid electrophoresis buffer widely used in biology, mainly used for agarose gel electrophoresis of DNA. When using TAE as the running buffer, the mobility of double-stranded linear DNA is faster during electrophoresis. When the fragment is larger than 13 kb, it is generally recommended to use TAE for electrophoretic separation. TAE is suitable for the recovery of DNA fragments. Due to the small buffer capacity of TAE, it is not recommended for long-term electrophoresis (such as overnight).
Protocol :
1. Measure about 600 ml of distilled water into the beaker, and place a magnetic stir bar in the beaker.
2. Place the beaker on a magnetic stirrer, slowly add the entire contents of 1 bag of TAE Instant Granules, and stir the solution until it is completely dissolved.
3. Add distilled water to the TAE solution in step 2, and dilute to 1 L, which is 1× buffer.
After being formulated into 1× solution, the pH value is 8.3±0.1@25℃.
