Rapid Running Buffer (hereinafter referred to as RRB) is white to almost white instant granules. Each bag of RRB instant granules can produce 1 L of 1×RRB buffer, which is easy to operate and easy to use. RRB buffer is a fast nucleic acid electrophoresis buffer, mainly used for DNA agarose gel electrophoresis. RRB buffer has the characteristics of strong buffering capacity and high resolution. It can effectively separate DNA fragments within 5 kb. With a certain concentration of agarose gel, it can support high-pressure fast electrophoresis at 300-350 V (15 V/cm). The routine electrophoresis time can be shortened from 30 minutes to about 10 minutes. This buffer can replace TBE running buffer for small molecular weight DNA agarose gel electrophoresis.

Protocol :

1. Measure about 600 ml of distilled water into the beaker, and place a magnetic stir bar in the beaker.
2. Place the beaker on a magnetic stirrer, slowly add the entire contents of 1 bag of RRB instant granules, and stir the solution until it is completely dissolved.
3. Add distilled water to the RRB solution in step 2, and dilute to 1 L, which is 1× buffer.
After being formulated into 1× solution, the pH value is 8.35±0.15@25℃.

Fast electrophoresis
The recommended concentration is 1.5% agarose gel, the voltage is adjusted to 300~350 V (15 V/cm), and the effective separation of 100~5000 bp DNA fragments can be completed within 10 minutes.
Note 1: When performing fast electrophoresis, the recommended agarose gel concentration is 1.5%. A gel concentration below this concentration may cause poor resolution of small fragments below 200 bp; gel concentration higher than this concentration for large fragments The band may be poorly distinguished;
Note 2: If the room temperature is too high or continuous electrophoresis will cause the temperature of the electrophoresis buffer to be too high, it is recommended to cool down before performing electrophoresis.
② Routine electrophoresis
1× RRB electrophoresis buffer can replace TBE for conventional electrophoresis operations to achieve effective separation of small molecular weight DNA fragments
