Rapid Blocking Buffer (TBS-T) instant granules are off-white to light yellow granules. Each bag can prepare 100 ml blocking solution, which is easy to operate. The main component is the fish gelatin protein component that has been carefully processed, and the protein content is 5% after being formulated into 1× working solution. This product is used in the antibody blocking step of Western Blot and ELISA. The TBS-T buffer (containing Tween20 detergent) version is provided.
The fish gelatin protein used in Rapid Blocking Buffer (TBS-T) is compatible with most proteins, providing excellent reaction performance and compatibility. It can be blocked within 15 minutes, shortening the user’s experimental time. It has been premixed with TBS-T for ease of use. In many Western Blot detection reactions, the presence of detergent Tween20 improves the efficiency of blocking.
Protocol :
Steps for preparation
1. Measure about 50 ml of distilled water into the beaker, and place a magnetic stir bar in the beaker.
2. Place the beaker on a magnetic stirrer, slowly add the entire contents of 1 bag of Rapid Blocking Buffer (TBS-T) instant granules, and stir the solution until it is completely dissolved.
3. Add distilled water to the Rapid Blocking Buffer (TBS-T) solution in step 2, and dilute to 100mL, which is 1× buffer
Steps for Blocking
1. After transferring the membrane, put the transfer membrane into the hybridization incubator, add 10-20 ml 1× Rapid Blocking Buffer (TBS-T), cover the surface of the carrier, and incubate on a tabletop horizontal shaker at room temperature for about 10 minutes.
Note: The blocking effect of this product for 10 minutes is significantly better than that of conventional BSA blocking for 1 hour. For antibodies with high background, you can try to extend the blocking time to 30-60 minutes.
2. The sealed membrane can be used for subsequent experiments such as primary antibody incubation.
