Safe Red DNA Stain has a large molecular weight (about 1240g/mol), which cannot penetrate cell membranes. So it is safe and non-toxic. The sensitivity of safe red DNA stain is higher than the traditional dye EB. Safe Red DNA Stain has the same spectral characteristics as EB and has a strong excitation wavelength in the range of 250-300 nm. It is suitable for imaging with a gel imager equipped with an ultraviolet light source and can perfectly replace EB.
Protocol :
1. Bubble dyeing method (recommended)
① Perform electrophoresis in accordance with conventional methods.
② Prepare 3× Safe Red DNA Stain staining solution. The specific method is: Dilute Safe Red DNA Stain 10,000× stock solution approximately 3,300 times to 0.1 M NaCl (for example: add 15 μl Safe Red DNA Stain 10,000× stock solution and 5 ml 1 M NaCl to 45 ml H2O) .
Note:
3× Safe Red DNA Stain staining solution can be prepared in large quantities and stored at room temperature in the dark until it is used up.
③ Put the gel carefully into a suitable container, slowly add enough 3× staining solution to immerse the gel, shake at room temperature for 10-20 min, and the optimal staining time will be slightly extended according to the thickness of the gel and the increase in agarose concentration. .
2. Paste dyeing method (same usage as EB)
① Add Safe Red DNA Stain safe nucleic acid dye during gel preparation (for example: add 5 μl of Safe Red DNA Stain 10,000× stock solution to every 50 ml agarose solution, and analogy with this ratio).
Note:
A. The amount of dyeing dyes in this method is relatively small, 500 μl of 10,000× stock solution can prepare about 100 pieces of 50 ml glue;
B. Safe Red DNA Stain has good thermal stability and can be added directly to the hot agarose solution. You can also choose to add Safe Red DNA Stain 10,000× stock solution to agarose powder and electrophoresis buffer, and then heat it in a microwave oven or other methods to prepare agarose gel.
② Perform electrophoresis in accordance with conventional methods.
